چکیده

Objective The primary objective of this study was to evaluate whether the polymorphism of poly (ADP-ribose) polymerase-1 (PARP-1) is involved as a potential risk factor in the development of acute renal injury in elderly Chinese patients with diabetes mellitus. Subjects and methods In this pilot study, diabetic patients of either gender (aged ≥65 years) with a confirmed diagnosis of acute renal injury and individuals with no clinical symptoms of acute renal injury were enrolled at Nanxishan Hospital of Guangxi Zhuang Autonomous Region, China. Genetic polymorphism of PARP-1 was assessed using polymerase chain reaction-restriction fragment length polymorphism assay. Cytokine levels (interleukin [IL]-6, IL-1β and tumor necrosis factor-alpha) were measured in the serum samples by sandwich enzyme-linked immunosorbent assay technique. Results A total of 130 Chinese patients with acute renal injury and 130 Chinese individuals with no clinical symptoms of acute renal injury were included. We found that the patients with GG genotype and carriers of the G and C alleles of PARP-1 were at high risk of developing acute renal injury. Moreover, del/ins polymorphism of the NF-κB1 gene was also found to be associated with acute renal injury. In addition, the levels of IL-6, IL-1β and tumor necrosis factor-alpha were significantly increased in patients with acute renal injury (p<0.05). Conclusion Our findings showed the involvement of PARP-1 polymorphisms in the development of acute renal injury in Chinese individuals. This study identified the involvement of two SNPs of PARP-1 (C410T and G1672A) in development of acute renal injury among Chinese diabetic patients. Also, increased expression of C and G alleles of PARP-1 can be considered as one of the potential risk factors for developing acute renal injury. Increased serum cytokine levels can be considered as one of the potential risk factors for developing acute renal injury.

The therapeutic potential of poly(ADP-ribose) polymerase inhibitors.

Poly(ADP-ribose) polymerase-1 (PARP-1) is a member of the PARP enzyme family consisting of PARP-1 and several recently identified novel poly(ADP-ribosylating) enzymes. PARP-1 is an abundant nuclear protein functioning as a DNA nick-sensor enzyme. Upon binding to DNA breaks, activated PARP cleaves NAD(+) into nicotinamide and ADP-ribose and polymerizes the latter onto nuclear acceptor proteins i...

The pathogenesis of diabetic complications: the role of DNA injury and poly(ADP-ribose) polymerase activation in peroxynitrite-mediated cytotoxicity.

Recent work has demonstrated that hyperglycemia-induced overproduction of superoxide by the mitochondrial electron-transport chain triggers several pathways of injury [(protein kinase C (PKC), hexosamine and polyol pathway fluxes, advanced glycation end product formation (AGE)] involved in the pathogenesis of diabetic complications by inhibiting glyceraldehyde-3-phosphate dehydrogenase (GAPDH) ...

Diabetes, microvascular complications, and cardiovascular complications: what is it about glucose?

Glycemic control is the primary mediator of diabetic microvascular complications and also contributes to macrovascular complications. A new study (see related article beginning on page 1049) reveals a previously unrecognized association between oxidant activation of poly(ADP ribose) polymerase (PARP) and upregulation of known mediators of glycemic injury. Inhibitors of PARP may have potential t...

Inhibition of poly(ADP-ribose) polymerase attenuates ischemic renal injury in rats.

The enzyme, poly(ADP-ribose) polymerase (PARP), effects repair of DNA after ischemia-reperfusion (I/R) injury to cells in nerve and muscle tissue. However, its activation in severely damaged cells can lead to ATP depletion and death. We show that PARP expression is enhanced in damaged renal proximal tubules beginning at 6-12 h after I/R injury. Intraperitoneal administration of PARP inhibitors,...

Involvement of poly(ADP-ribose) polymerase-1 in development of spinal cord injury in Chinese individuals: a Chinese clinical study

Objective We aimed to evaluate whether the polymorphism of poly(ADP-ribose) polymerase-1 (PARP-1) is involved as potential risk factor in the development of spinal cord injury (SCI) among Chinese individuals. Patients and methods Patients with a confirmed diagnosis of SCI (other than traumatic injury) and healthy individuals with no clinical symptoms of SCI were enrolled at Spinal Cord Injury...
